Every immune recipe starts with the same two staples.

The innate immune system stocks two fundamental ingredients. PAMPs — pathogen-associated molecular patterns — are molecules derived from microbes, the unmistakable signature of a foreign intruder like bacterial LPS. DAMPs — damage-associated molecular patterns — are host-derived, released by stressed or dying cells to signal injury.

Think of them as your two baking staples: PAMP flour (is this thing foreign and pathogen-like?) and DAMP sugar (is real harm actually happening?). One tells you what it is; the other tells you whether it’s hurting you. A good filter needs both in the bowl.

The secret is the blend, not any single dose.

Here’s the immune system’s masterstroke: PAMPs and DAMPs don’t act alone — they synergize. Research shows the two classes must combine to permit full secretion of pro-inflammatory cytokines like IL-1β. Flour without sugar, sugar without flour: neither rises on its own.

III · THE TWO-SIGNAL RULERequire a pinch of context

A great filter waits for confirmation before it fires.

The body insists on corroboration. Under the two-signal model, initiating a response often requires both a microbe-derived ligand (signal one) and additional contextual information that accompanies a real threat (signal two). Your filter should bake the same way: one suspicious marker is dough, not a verdict. Demand a confirming pinch of context — pattern, history, corroborating reports — before the post comes out of the oven flagged.

Blend, don’t solo

PAMPs and DAMPs synergize for a full response. Combine signals; never let one feature alone trip the filter.

The whole recipe in one act of seasoning.

Every bake weighs the danger signals (PAMP flour and DAMP sugar — foreign and harmful) against the safe signals (SAMP salt — markers that vouch for normalcy). The aim is to react to genuine harm, not mere foreignness — an unusual post is novel, not necessarily spicy.

And one stray ingredient shouldn’t change the whole loaf. Let signals accumulate and synergize past a threshold before flagging, and set the dial so only the truly spicy posts rise to the top.

When the recipe overshoots

Aberrant or excessive PRR activation drives inflammatory and autoimmune disorders — the system attacking healthy tissue. And accumulating DAMPs can spark a vicious circle of runaway inflammation.
